I made a bootable usb stick and used it for a while. I then messed with one of the .lzm files in the "base" directory. I don't think that would have messed up anything but... Now my usb stick is apparently a read-only filesystem. I can't delete or put anything on it, even as root. What could I have done to cause this? Or is it a defective usb stick? Would reformatting it help?

are you using this to boot?

issue the mount command and see if it's mounted read-only

probably just need to remount it in rw mode

Hmm... I plugged it in again (to try and see if it was mounted read-only) but before I checked, I tried to delete something and it worked.  Huh Maybe it just needed a good safely-remove-and-plug-back-in.

I had that happen once.  For some reason the usb icon on my desktop didn't go away from an earlier use.  I opened the desktop, deleted the icon, and it worked fine thereafter.
